As we mentioned eaasier, trying to actually measure the voltage across the wire will be very difficult, which is why we strongly recommend using a thermocouple interface chip. The best we've seen so far is the MAX6675, which is a SOIC package.
Thermocouples are two wires soldered together, the chip reads the voltage difference between the two. One end is negative (K-type, made of Al-Ni alloy) and the other end is positive (Ni-Cr alloy). They can be distinguished by color. In general, red is negative and yellow is positive.
CLK (Clock) is an input to the MAX6675 (output from the controller) that provides another bit of data when indicated.
DO (Data Out) is the output of the MAX6675 (input to the controller) which outputs each bit of data.
CS (Chip Select) is input to the MAX6675 (output from the controller) which controls when the chip reads the thermocouple and outputs more data
